Not cities. It moves a couple miles from the VAB (=Vehicle Assembly Building, the largest building south of the Pentagon) to the launch pad. A special "crawler" vehicle carries it there.

It also travels on the back of a specially modified Boeing 747 when it Lands at Edwards AFB, CA or any other diversion site. All spce shuttles traveled to Florida from Palmdale, CA in this manner after they were built.
