Patiently. If you are referring to someone that is intoxicated they will just need repeating in a calm voice no matter how much they over react.

In a calm and proffesional manner. If the unfit refers to drugged or drunk, then they should not get on the plane so you have to be set to deal with a lot of anger and stress.
If they are on the flight in mid air then they should be dealt with clamly and if possible re-seated away from other passengers.
